What it is
Spermidine is a polyamine, a small positively charged molecule present in all living cells and involved in DNA stability, cell growth and protein synthesis. It was first identified in semen, which explains the name, but is widespread throughout the body and in food.
Rich dietary sources include wheat germ, aged cheeses, mushrooms, soybeans and natto. Wheat germ extract is the main commercial supplement source, though isolated spermidine trihydrochloride is also used.
Cellular spermidine content declines with age, which parallels the pattern seen with NAD+ and forms the basis of interest in it as a longevity compound. A practical point worth noting: wheat germ extract products contain gluten, which matters for a meaningful share of customers.
How it works
The most studied mechanism is induction of autophagy, the process by which cells break down and recycle damaged components. Autophagy declines with age, and its restoration is a central target in ageing research. Spermidine appears to trigger it through inhibition of a specific acetyltransferase.
Spermidine also stabilises DNA and RNA structures, participates in the hypusination of eIF5A, a modification required for translation of certain proteins, and influences mitochondrial function.
An open question is how much orally administered spermidine reaches cells intact, given that polyamines are extensively metabolised in the gut and that gut bacteria both produce and consume them. Whether supplementation raises tissue spermidine meaningfully is not firmly established.
What the evidence shows
Animal research is the strongest part of the literature, with reported lifespan extension in yeast, flies, worms and mice, alongside cardiac and cognitive findings in rodents. This body of work is genuinely interesting.
Human evidence is largely observational. Population studies have reported associations between higher dietary spermidine intake and lower mortality, which is suggestive but subject to the usual confounding: people eating more wheat germ, mushrooms and fermented foods differ in many ways.
A small number of intervention trials, mainly using wheat germ extract in older adults, have reported modest cognitive findings and good tolerability. Sample sizes are small and durations short. The compound is scientifically credible and the human case remains unproven, which is a fair place to leave it.
Dosing
Supplemental doses are small, typically 1-6 mg of spermidine daily. Human trials have generally used wheat germ extract supplying around 1-6 mg.
Dose against declared spermidine content, not extract weight, as extracts vary considerably in concentration.
Dietary intake in a typical Western diet is estimated in the region of 10-15 mg daily, meaning supplements add a modest increment rather than a transformative one. Eating more wheat germ, mushrooms and aged cheese is a legitimate alternative route, and worth saying plainly. Usually taken in the morning with food.
