What it is
L-histidine is an essential amino acid, though it was classified as conditionally essential for some time because adults appear able to manage short periods without it. It is found in meat, fish, poultry, dairy, eggs and wholegrains.
It has two roles worth knowing. It is the precursor to histamine, the mediator involved in allergic responses, gastric acid secretion and wakefulness. And it combines with beta-alanine to form carnosine, the intramuscular buffer.
That second role explains why histidine is rarely supplemented. Carnosine synthesis is limited by beta-alanine availability, not histidine, since muscle histidine is generally plentiful. Supplementing histidine to raise carnosine addresses the wrong bottleneck.
How it works
Histidine's imidazole side chain gives it unusual chemistry, able to gain or lose a proton at physiological pH. This makes it an effective buffer in its own right and central to the function of many enzymes, where histidine residues sit at active sites.
Histidine decarboxylase converts it to histamine, which acts on multiple receptor types affecting immune response, stomach acid production and arousal in the brain.
Carnosine synthase joins histidine with beta-alanine to form carnosine. Because beta-alanine is the limiting substrate, raising histidine has little effect on carnosine content, which is a clear example of why identifying the actual bottleneck matters in supplementation.
What the evidence shows
There is little evidence supporting histidine supplementation in healthy people eating adequate protein, and no established performance benefit. Trials adding histidine to beta-alanine have not shown additional carnosine loading.
Lower histidine status has been observed in some groups under medical care, and there is limited research on supplementation in those settings. That work does not transfer to a healthy person eating plenty of protein.
A small body of work on metabolic markers in overweight women has reported favourable findings, but this is preliminary. Histidine is best understood as physiologically important and supplementally uninteresting for most people.
Dosing
Supplemental doses, where used, range from 500 mg to 4 g daily. There is no widely established protocol because there is no widely established reason to take it.
If the goal is raising muscle carnosine, beta-alanine is the ingredient that works, and adding histidine to it does not improve the result.
EAA products contain histidine as one of the nine essentials, which is the most sensible way to receive it if you want the full spectrum. Isolated histidine supplementation is difficult to justify for anyone eating adequate protein.

Safety and side effects
Well tolerated at dietary and modest supplemental intakes. High doses have been associated with headache, nausea and weakness in some reports.
Histidine is the precursor to histamine, so anyone who reacts badly to histamine-rich foods, or who is prone to strong allergic responses, may find supplemental histidine aggravates that. This is the main practical caution.
