Undenatured Type II Collagen

Also known as: UC-II, UCII, Native type II collagen, Chicken sternum collagen

Joint & ConnectiveGood evidence

Collagen taken at 40 mg rather than 10 g, working through an immune mechanism rather than by supplying building blocks. Not a smaller dose of collagen peptides — a completely different thing.

Typical dose

40.0–40.0 mg per day

40 mg once daily on an empty stomach, usually before bed

What it is

Undenatured type II collagen is collagen that has deliberately not been broken down. It is processed at low temperature to keep its triple helix structure intact, which is the opposite of how collagen peptides are made.

It is sourced from chicken sternum cartilage, which is naturally rich in type II collagen, the form found in joint cartilage rather than skin.

The dose is the giveaway. Collagen peptides are taken at 10 g or more; this is taken at 40 mg, roughly 250 times less. That is not a weaker version of the same product. If a supplement claims to be UC-II and lists grams rather than milligrams, something is wrong.

How it works

The mechanism is oral tolerance, an immune process. Small amounts of an intact protein passing through specialised immune tissue in the small intestine teach the immune system to recognise it as harmless rather than as something to attack.

Applied to joint cartilage, the proposal is that this trains the immune system to stop attacking the body's own type II collagen, reducing the immune contribution to joint wear.

Two things follow from this. The collagen must remain intact, which is why processing matters. And the dose must be small, because oral tolerance works with tiny quantities and larger amounts can defeat it. This is one of the few supplements where more would be actively worse.

What the evidence shows

Trials in adults with joint discomfort have reported improvements in comfort, stiffness and physical function over 90 to 180 days, and some have reported better results than glucosamine and chondroitin combined.

There is also research in healthy active adults with exercise-related knee discomfort, reporting improved range of motion and reduced discomfort during activity, which is more directly relevant to a training audience.

Trial sizes are modest and industry funding is common, the usual caveats. But the mechanism is distinctive, the dose is coherent with it, and the evidence compares reasonably well against the far more heavily sold glucosamine and chondroitin.

Dosing

The dose is 40 mg daily, which typically supplies around 10 mg of actual undenatured type II collagen. This figure is consistent across essentially all the research. Do not take more. Oral tolerance depends on small quantities, and larger doses may work against the mechanism rather than improving on it. Take on an empty stomach, generally before bed, which is what the trials used. Effects accrue over 90 to 180 days, so this is a slow supplement and judging it after a month is premature. It is not a substitute for collagen peptides, which do a different job at a vastly different dose.

Safety and side effects

Undenatured type II collagen is well tolerated at the small doses used, with few adverse effects reported in trials. Mild digestive upset is the occasional complaint.

It is derived from chicken, so anyone with a poultry allergy should avoid it, and it is not suitable for vegetarians or vegans.

Because the mechanism involves modulating an immune response, speak to your doctor before use if your immune system is being managed.

Who should avoid it

Derived from chicken; avoid with poultry allergy and not suitable for vegetarians or vegans. Speak to your doctor before use if your immune system is being managed, given the immune-based mechanism. Do not exceed the 40 mg dose. Not established in pregnancy or breastfeeding.
