What it is

Ecdysterone, more precisely 20-hydroxyecdysone, is a steroid hormone found in insects and in a range of plants, where it is thought to serve a defensive function. Dietary sources include spinach, quinoa and asparagus, though at concentrations far below supplemental doses.

Commercial material is typically extracted from Cyanotis arachnoidea, Rhaponticum carthamoides or Ajuga turkestanica. Extract quality varies dramatically, and independent testing of retail products has repeatedly found actual ecdysterone content well below label declaration.

Despite being structurally a steroid, ecdysterone does not bind the androgen receptor, which is the basis for describing it as non-androgenic. It should not be conflated with anabolic androgenic steroids or prohormones, which act through entirely different mechanisms and carry entirely different risk profiles.

How it works

The most cited proposed mechanism is activation of the oestrogen receptor beta pathway, which in preclinical models triggers downstream signalling associated with protein synthesis. This is distinct from androgen receptor activation and does not produce oestrogenic effects in the way that term might suggest.

Animal and cell studies have reported increased protein synthesis, enhanced myotube growth and effects on the Akt/mTOR pathway. Some work also suggests improvements in glucose handling and reduced protein degradation.

A persistent difficulty is bioavailability. Ecdysterone appears to undergo rapid metabolism and clearance, and oral bioavailability estimates in mammals are low. Reconciling the potency observed in cell culture with what an oral dose can plausibly deliver to muscle tissue remains an unresolved gap in the case for this compound.

What the evidence shows

Human evidence is thin. The most influential study is a ten-week trial in resistance-trained men which reported greater increases in lean mass and strength in supplemented groups compared with placebo. It attracted considerable attention and was a factor in subsequent anti-doping interest.

That trial has not been convincingly replicated at scale. Earlier human work reported no meaningful effect, and the overall body of human data remains small, with modest sample sizes and inconsistent extract standardisation between studies. Building confident conclusions on a single positive trial is not sound practice, which is why the grading here is limited rather than moderate.

The product quality issue compounds this. Where independent analysis finds retail products containing a fraction of their declared ecdysterone, consumer experience becomes an unreliable guide to whether the compound itself works.

Dosing

Supplemental protocols commonly use 200-1,000 mg daily of ecdysterone, usually split across two servings. The influential human trial used doses at the higher end of that range. Dose against declared ecdysterone content, not milligrams of plant extract. A 500 mg serving of a 5 per cent extract supplies 25 mg of ecdysterone, which is not comparable to 500 mg of standardised material. This distinction is where most consumer confusion in the category originates. Typically run in blocks of 8-12 weeks alongside resistance training, since the proposed effects depend on a training stimulus.

Safety and side effects

Reported side effects in the available human trials have been minimal, and ecdysteroids have substantial dietary exposure through common foods such as spinach and quinoa. No hormonal suppression has been demonstrated, which is consistent with the absence of androgen receptor binding, and no post-cycle therapy is required.

The genuine caveat is that long-term human safety data at supplemental doses is sparse. Absence of reported harm in short trials with small samples is not the same as an established safety profile, and this should be stated plainly rather than glossed as safe.

Product quality also functions as a safety issue in its own right. Poorly controlled botanical extracts carry risks of adulteration and contamination independent of the target compound.
