What it is
D-aspartic acid is the D-isomer of aspartic acid, a mirror image of the L-form found in proteins. Unlike most D-amino acids, which are biologically inert in humans, D-aspartic acid accumulates in specific tissues including the pituitary, testes and brain, which is what prompted interest in it.
It is not obtained meaningfully from diet. Supplements typically supply D-aspartic acid or its sodium salt, in doses of 3 g.
DAA is a useful case study in how supplement categories form. A single promising study produced an entire product category, and the subsequent research that failed to support it received a fraction of the attention.
How it works
The proposed mechanism operates at two levels. In the hypothalamus, D-aspartic acid is thought to stimulate release of gonadotropin-releasing hormone, which prompts the pituitary to release luteinising hormone.
In the testes, it is proposed to act directly on Leydig cells to support testosterone synthesis, with reported effects on the enzymes involved in steroidogenesis.
The mechanism is coherent on paper, which is why the ingredient was taken seriously. What the human trials showed is that a plausible mechanism does not guarantee a real effect, particularly in men whose hormonal axis is already functioning normally.
What the evidence shows
The founding study reported a meaningful increase in testosterone in untrained men over twelve days, and this single result generated the entire category.
Subsequent trials told a different story. A study in resistance-trained men found no increase in testosterone and no change in body composition or strength. Another trial testing both 3 g and 6 g found no benefit at the lower dose and, at the higher dose, a decrease in testosterone.
Reviews have generally concluded that DAA does not raise testosterone in resistance-trained men with normal hormonal status. Any effect appears confined to men with impaired baseline function, which is a much narrower proposition than how it is sold.
Dosing
The conventional dose is 3 g daily, taken in the morning, and this is what the research used.
Going higher is specifically inadvisable. The trial testing 6 g daily found lower testosterone at that dose than at baseline, so the usual assumption that more might work better appears to be wrong here.
Protocols typically run two to four weeks with breaks, though given the evidence this is a matter of convention rather than of demonstrated benefit. If the goal is testosterone in a healthy trained man, the honest answer is that this is not the ingredient for it.

Safety and side effects
Generally well tolerated at the standard dose. Reported effects include headache, irritability, nervousness and digestive upset, mostly mild.
The finding that a higher dose reduced testosterone is worth treating as a safety consideration rather than just an efficacy one, since it suggests the compound can act against the intended direction at higher intakes.
D-aspartic acid is also an NMDA receptor agonist, and there has been some discussion in the literature about excitotoxicity at high doses, though this has not been demonstrated as a practical concern in humans. Long-term safety data is limited.
Who should avoid it
Not suitable for women, particularly during pregnancy or breastfeeding. Not suitable for under-18s. Speak to your doctor before use if your hormones are being monitored or managed, or if you take regular medication. Do not exceed the conventional dose, given evidence that higher intakes may reduce testosterone.
