What it is

Carnosine is a dipeptide made of beta-alanine and histidine, stored in skeletal muscle where it acts as an intracellular buffer against the acidity produced during hard exercise.

It is found in meat and fish, particularly red meat, which is why vegetarians tend to have lower muscle carnosine.

Taking carnosine to raise muscle carnosine seems like the direct route and is not. An enzyme in the blood called carnosinase breaks ingested carnosine straight back into beta-alanine and histidine before it reaches muscle. You end up supplying beta-alanine anyway, just at higher cost and lower efficiency.

How it works

Within muscle, carnosine buffers hydrogen ions produced by anaerobic glycolysis, moderating the pH drop that interferes with contraction. Its buffering range is well suited to the pH encountered in working muscle.

Muscle carnosine is synthesised inside the muscle cell by carnosine synthase, using beta-alanine and histidine. Beta-alanine availability is the limiting factor, which is the entire basis for supplementing it.

Because circulating carnosinase degrades ingested carnosine rapidly, oral carnosine does not deliver intact dipeptide to muscle. Understanding this one enzyme explains why the whole product category is misconceived for that purpose.

What the evidence shows

The evidence that beta-alanine raises muscle carnosine and improves high-intensity performance is strong. The evidence that oral carnosine does the same is not, and the mechanism explains why.

There is separate research interest in carnosine's antioxidant activity and its ability to bind reactive carbonyl compounds, largely preclinical and unrelated to the buffering role.

Zinc carnosine is a genuinely different product with its own small evidence base concerning the gut lining, where it acts locally rather than systemically. It should not be confused with carnosine bought for performance.

Dosing

Products typically supply 500 to 1,000 mg daily. If your goal is raising muscle carnosine for performance, this is the wrong purchase and no dose fixes that. Beta-alanine at 3.2 to 6.4 g daily is the evidence-based route, and it is cheaper. That is the practical recommendation this page exists to make. Zinc carnosine, used for gut lining support at around 75 mg twice daily, is a separate product with a separate rationale, and buying one when you meant the other is an easy mistake given the naming.

Safety and side effects

Carnosine is well tolerated and is a normal dietary component of meat and fish. Adverse effects at supplemental doses are uncommon and mild.

Because ingested carnosine is broken down into beta-alanine, some people experience the same tingling sensation that beta-alanine causes, particularly at higher doses.
