What it is

Apigenin is a flavone, a subclass of flavonoid, present across a wide range of plant foods. The richest common sources are dried parsley, chamomile flowers and celery, with smaller amounts in oregano, thyme and citrus. Chamomile tea is the most familiar dietary route in the UK.

In plants it occurs largely as glycosides such as apiin, which must be cleaved before absorption. Supplements typically supply the aglycone form directly, either isolated or as a standardised chamomile extract.

Bioavailability is a genuine limitation. Apigenin is poorly water soluble and undergoes substantial first-pass metabolism, so plasma concentrations achieved from oral supplementation are far below the concentrations used in most cell-based research. This gap is the single most important caveat when interpreting the literature.

How it works

The most cited mechanism is weak binding at a site on the GABA-A receptor, the main calming receptor system in the brain. Apigenin's affinity there is orders of magnitude lower than that of compounds designed to act on it, producing at most a mild modulatory effect rather than genuine sedation.

Apigenin also inhibits CD38, an enzyme that consumes NAD+. This has generated interest in the longevity and cellular energy space, since raising NAD+ availability is a target of that category. The relevance at achievable oral doses in humans is unestablished.

Additional documented activities include effects on hormone-converting enzymes in laboratory models, which underpins its occasional appearance in hormonal support formulas, and a broad range of antioxidant and anti-inflammatory signalling effects typical of flavonoids. Almost all of this work is in vitro or in animals.

What the evidence shows

There are very few human trials of isolated apigenin. The clinical evidence most often cited in support of it actually comes from chamomile extract trials, where apigenin is one constituent among many. Those trials have reported modest improvements in sleep quality measures, but attributing the result to apigenin specifically is an inference rather than a finding.

The preclinical literature is large and consistent in showing calming effects in rodent models, alongside a substantial volume of cell-based laboratory work. Neither translates automatically to a supplement dose in a healthy human.

The honest position is that apigenin is mechanistically interesting, has a long history of safe dietary exposure, and has plausible but unproven benefits for sleep and relaxation at supplemental doses. Anyone claiming more than that is going beyond the data.

Dosing

Common supplemental range is 50-100 mg daily, though this range derives from product convention rather than dose-ranging trials. Some sleep protocols use up to 200 mg. Usually taken in the evening, 30-60 minutes before bed, on the basis of the proposed GABAergic mechanism. Given poor solubility, taking it with a source of dietary fat is a reasonable practical step, though formal bioavailability data supporting this in humans is limited.

Safety and side effects

Apigenin has extensive dietary exposure through common foods and chamomile tea, and no significant safety signal has emerged at supplemental intakes. Reported effects are mild and infrequent, principally drowsiness and occasional digestive upset.

The main practical consideration is additive drowsiness. Because it acts, however weakly, on the brain's calming receptor system, combining it with alcohol or anything else that makes you sleepy may compound the effect.

People sensitive to chamomile, ragweed, daisies or related plants may react to chamomile-derived preparations.
