What it is
L-citrulline is a non-protein amino acid named after Citrullus, the watermelon genus from which it was first isolated. It is produced in the body as part of the urea cycle and obtained in small amounts from watermelon and other cucurbits.
Its defining property is counterintuitive: citrulline raises plasma arginine more effectively than an equivalent dose of arginine does. It bypasses the arginase enzyme that degrades oral arginine on first pass, travels to the kidneys and is converted to arginine there, entering circulation intact.
Two forms dominate. Pure L-citrulline is straightforward. Citrulline malate combines citrulline with malic acid, usually at a 2:1 ratio, meaning 8 g of citrulline malate supplies roughly 5.3 g of citrulline. Some products use 1:1. Since the ratio is frequently omitted from labels, this is one of the more common sources of accidental underdosing in pre-workout formulas.
How it works
Citrulline converts to arginine, which nitric oxide synthase uses to produce nitric oxide. Nitric oxide relaxes vascular smooth muscle, increasing blood flow to working tissue and producing the perceptible fullness described as pump.
Beyond vasodilation, citrulline participates in ammonia clearance through the urea cycle. Ammonia accumulates during intense exercise and contributes to fatigue, so improved clearance offers a second, independent route to better work capacity.
The malate component is often credited with a role in the citric acid cycle as an energy intermediate. This is plausible but considerably less well substantiated than the citrulline effects, and should not be the reason for choosing the malate form.
What the evidence shows
The most consistent findings concern muscular endurance in resistance training. Several controlled trials have reported increased repetitions to failure across multiple sets following acute citrulline malate ingestion, and meta-analyses have generally supported a modest benefit.
Reduced muscle soreness in the days after training has also been reported in several trials. Effects on maximal strength and on power in single efforts are less consistent, which fits a mechanism operating through blood flow and metabolite clearance rather than force production.
Evidence that citrulline raises plasma arginine more effectively than arginine is solid and repeatedly demonstrated. Cardiovascular research, including work on blood pressure and endothelial function, is a smaller but reasonably encouraging strand. Underdosing is widespread, and many trials showing no effect used doses well below the effective range.
Dosing
Effective dosing is 6-8 g of pure L-citrulline, taken 30-60 minutes before training. If using citrulline malate at 2:1, that means roughly 9-12 g of the blend to deliver an equivalent citrulline dose.
This is where most pre-workout products fall short. A formula listing 3 g of citrulline malate supplies around 2 g of citrulline, well below what trials used. Check the ratio, and if it is not stated, assume the worst.
Daily use is not required for the acute pump effect, though cardiovascular research has generally used consistent daily intake over weeks. Citrulline is quite sour in malate form; pure citrulline is more neutral.

Safety and side effects
Citrulline has an excellent tolerability record. Unlike arginine, it does not commonly cause gastrointestinal distress even at effective doses, which is a practical advantage that often decides the choice between them.
Reported effects are mild and infrequent: occasional stomach discomfort, and headache in some users, likely related to vasodilation. Trials at doses up to 15 g have not reported significant adverse effects.
Because it promotes vasodilation, it may modestly lower blood pressure, relevant to anyone on antihypertensive medication. It should not be combined with nitrate medication or PDE5 inhibitors without advice.
Who should avoid it
Do not combine with any medication used to widen blood vessels or manage blood pressure without medical advice. Speak to your doctor before use if you take regular medication, or if you have been told your kidneys are not working normally. Not established for use in pregnancy or breastfeeding.
